The MESACUP-2 TEST anti-TPO is the quantitative enzyme-linked immunosorbent assay (ELISA) for the detection of anti-Thyroid Peroxidase (TPO) antibodies in human serum.
Background:
Thyroid Peroxidase (TPO) is one kind of hem protein, which is located in microsome fraction in thyroid cell and involved in cuppling of tyrosine residues and iodization in thyroglobulin molecule in the process of synthesizing thyroid hormone. In thyroid microsome fraction of autoimmune thyroid disease patients, it has been known anti-microsome antibodies are present with high frequency, and in recent research, it is shown that major antigen of anti-microsome antibodies are TPO. Anti-TPO antibodies are a complement fixing antibodies, has cytotoxicity and is known to show high value in the serum of autoimmune thyroid disease patient such as Hashimoto diseases or basedow’s diseases. So detecting anti-TPO antibodies in the serum is widely practiced together with anti thyroglobulin antibody as diagnosis and indication of treatment of thyroid disease.
The MESACUP-2 TEST anti-TPO is for detecting anti-TPO antibodies in the serum with ELISA (Enzyme-Linked Immunosorbent Assay).
Principle:
The MESACUP-2 TEST anti-TPO measures anti-TPO antibodies present in the serum by ELISA. Standards and patient sera are added to microwell coated with Tg antigens, allowing anti-Tg antibodies to react with the immobilized antigen (Sample incubation). After wash to remove any unbound serum proteins, horseradish peroxidase conjugated anti human IgG polyclonal antibody (goat) is added and incubated (Conjugate incubation). Following another washing step, the peroxidase substrate is added and incubated for an additional period of time (Substrate incubation). Acid solution is then added to each well to terminate the enzyme reaction and to stabilize the color development. The assay can be quantified by measuring the reaction photometrically and plotting the results.
