The MESACUP Cardiolipin TEST is the quantitative enzyme-linked immunosorbent assay (ELISA) for the detection of anti Cardiolipin antibodies in human serum.
Background:
As well as biologically false positive serologic tests for syphilis (BFP) and lupus anticoagulant (LAC), anti-Cardiolipin antibodies are one of the anti-phospholipid antibodies. This antibody reacts with cardiolipin or complex composed of cardiolipin and cofactor (ex.β2 glycoprotein 1) in human or animal sera. These antibodies are associated with thrombosis, organ infarct, recurrent fetal loss and thrombocytopenia. To describe patients who show the common features to these diseases, Hughes, Harris et al. introduced the new concept termed "anticardiolipin syndrome" in 1986. At present, it is widely recognized as "anti-phospholipid antibody syndrome". Anti-Cardiolipin antibodies can be measured by RIA or ELISA method. Since there were various methods and reference samples used to measure these antibodies in many laboratories, Harris’s ELISA method were standardized in the International Work shop of 1986.
The MESACUP Cardiolipin TEST is for measuring with high sensitivity, anti-Cardiolipin antibodies present in the sera by ELISA.
Principle:
The MESACUP Cardiolipin TEST measures anti-Cardiolipin antibodies present in the serum by ELISA. Standards and patient sera are added to microwell coated with Cardiolipin, allowing anti-Cardiolipin antibodies to react with the immobilized antigen (Sample incubation). After wash to remove any unbound serum proteins, horseradish peroxidase conjugated anti human IgG monoclonal antibody is added and incubated (Conjugate incubation). Following another washing step, the peroxidase substrate is added and incubated for an additional period of time (Substrate incubation). Acid solution is then added to each well to terminate the enzyme reaction and to stabilize the color development. The assay can be quantified by measuring the reaction photometrically and plotting the results.
