When a protein to be investigated is artificially expressed in cells or tissues, "tags" (labels) composed of peptides or fluorescent proteins may be fused to the target protein for use as an indicator of expression. Dynamics of the target protein can be analyzed by western blotting, immunoprecipitation, and cell staining using specific antibodies that recognize these tags. MBL has a large choice of products, such as high-performance antibodies against fluorescent proteins, including GFP and RFP, and various epitope tags, including Myc, His, HA, V5, and DDDDK. MBL also provides tag-fused protein purification kits using these antibodies. These kits has the advantage of purifying proteins without damage.

Autophagy is a bulk self-degradation system involving the degradation of intracellular organelles and abnormal proteins that accumulated in the cells. The degradation products are reutilized for biosynthesis. Autophagy defects in mammalian are involved in various diseases such as neurodegenerative diseases, inflammatory bowel diseases, and cancers. Especially, in cancer, clinical trials are carried out using autophagy inhibitors. However, molecular mechanisms of autophagy are not fully understood and importance of basic research is pointed out.

In collaboration with leading autophagy researchers, MBL develops and sells high quality antibodies. Especially, anti-LC3 antibodies, essential tools for monitoring autophagy, receive high reputations and have been used in many research articles. We also offer high quality antibodies against p62 and NRF2, which are indicated to be involved in canceration. Recently, sampler set of autophagy-related antibodies also joined our product lineup.

RiboCluster Profiler™

RNA-binding proteins (RBPs) play an important role in regulating post-transcriptional gene expression, including mRNA splicing, nuclear export, intracytoplasmic localization, stability, and control of translation efficiency.

"RIP-Assay Kit" and "RIP-Certified Antibody" can be used for comprehensive analysis of functionally related mRNAs by RNP immunoprecipitation (RIP) of a complex of RBP and mRNA (mRNP) with an antibody against RBP. "RIP-Assay Kit for microRNA" can isolate miRNA and related mRNA using RIP technology. "RiboTrap Kit" can isolate proteins which bind mRNA of interest. These products attract attention as useful tools for researching posttranscriptional regulation mechanisms of genes that have been difficult to elucidate by conventional gene and/or protein expression analyses.

Epigenetic regulation of gene expression

Epigenetics refer to mechanisms of gene expression regulation by information other than DNA sequence such as chemical modifications. Epigenetics now get much attention since epigenetic regulation is revealed to be involved in development and diseases such as cancer. The chemical modifications that influence gene expression include DNA methylation, histone methylation and histone acetylation. MBL launched a kit for methylated DNA recovery "MethylHunter MBD1-based Methylated DNA Enrichment Kit" in 2011, and "MethylHunter MBD1- based Methylated DNA Enrichment Kit 2" which improves convenience of usage and achieves higher methylated DNA concentration in 2013. We also sells many self-developed antibodies as well as ChIP-grade antibodies against modified histones developed by MAB Institute, Inc.

Mouse female cells were immunostained by several antibodies that were directly labeled with different fluorescent dyes. Anti-H3K9me3 antibody strongly labeled constitutive heterochromatin and Anti-H3K27me3 antibody strongly labeled facultative heterochromatin.

MHC tetramerMajor histocompatibility complex (MHC) is a collective term used to refer to genes that encode proteins involved in the recognition of self and non-self antigens. MHC is involved in immune response such as elimination of virus-infected cells and tumor cells and rejection of the transplanted organs.

MHC molecules are subdivided into class I and II molecules. Class I molecules are expressed in most of cells and platelets, and class II molecules are expressed in antigenpresenting cells such as dendritic cells and macrophages. Immune response that occurs in our body involves T cells, and T cell receptors (TCR), which are expressed on each T cell surface, function by specifically binding to MHC that binds to antigen peptides (MHC/ peptide complex).

MHC tetramers are tetramerized MHC/peptide complex that are labeled with fluorescence molecules. MHC tetramers can be used for the isolation as well as detection of antigenspecific T cells. Thus, these tetramers enable detailed functional analysis and phenotyping of antigen-specific T cells. MHC tetramer reagents are now widely used as monitoring tool of T cell immune response in research and development of infection, cancer vaccine therapy, cellular immune therapy, transplantation immunity, and autoimmune diseases. MBL exclusively develops and produces Class I and Class II MHC tetramers, the novel tools for immunological studies.

-Kit for iPS cell induction without damages on chromosomes in cells-

"CytoTune®-iPS," a Sendai virus vector-based iPS cell induction kit developed by ID Pharma Co., Ltd.*, is composed of four Sendai virus vectors (SeV vectors), each carrying one of the Yamanaka reprogramming genes (OCT3/4, SOX2, KLF4, c-MYC) required for efficient reprogramming to induced pluripotent stem cells (iPS cells), and has been shown to induce iPS cells from human somatic cells. Because of characteristics of the SeV vector, this product enables the induction of chromosomal damage-free iPS cells and removal of the vector and the reprogramming genes from iPS cells.

Considering the influence on the environment and safety, the SeV vectors used in CytoTune®-iPS were modified to produce or release no infectious viral particles. This kit consists of a monopolistic technology based on a patent of ID Pharma related to the Sendai virus vector, and a monopolistic technology of iPS Academia Japan, Inc. related to the reprogramming gene.

Fucci: Fluorescent ubiquitination-based cell cycle indicator

FucciA fluorescent ubiquitination-based cell cycle indicator (Fucci), developed by RIKEN in 2007, is a fluorescent probe that enables real-time observation of cell cycle progression. Two different fluorescent proteins developed by Amalgaam, a green-colored monomeric, "Azami-Green1" (mAG1) and an orange-colored monomeric, "Kusabira-Orange2" (mKO2), are utilized in the Fucci system. Using the system, we can image spatio-temporal cell cycle patterns in various life phenomena such as cell proliferation and differentiation, and behavior of cancer cells which cannnot be analyzed by conventional methods.
* This product is licensed to use an invention by the independent administrative agency RIKEN and the incorporated foundation Tokyo Metropolitan Institute of Medical Science.

Vectors targeting intracellular organelles

Plasmids for expressing fluorescent proteins to specifically label organelles are designed to express individual fluorescent proteins in a target sitespecific manner using signal peptides. This allows easy labeling of each organelle with a preferred color.

Antibodies against regulatory T cells

Regulatory T-cellsRegulatory T cells (Treg) function as suppressors of the immune system by inhibiting the function of activated T cells. These cells mediate immunosuppression involved in various immune reactions such as autoimmunity, tumor immunity, and transplantation immunity. The onset of cancer and allergy is related to the balance between activated T cells and Treg.

MBL provides Treg-specific antibodies and antibodies that block mouse FR4 and GITR in vivo.

Mucosal immunity-related antibody

Mucosal immunityThe intestinal tract is an organ covered with mucosa, being exposed not only to foods as nutrition source, but also to various pathogens. Microfold cells (M cells) scattered on the inner wall of the intestine play a role as a "gate" that uptakes various substances from digested foods and transfers these substances to immune cells present immediately below the mucosa. If the immune cells recognize the transferred substances as "foreign substances," the cells induce a response for eliminating them. However, because no molecular markers for the M cells had been found, analysis of the mechanism to uptake substances by the M cells had not progressed.

In 2008, a glycoprotein, GP2, was discovered as a molecular marker that is specific for the M cells. GP2 is the first marker molecule for M cells that is common to man and animals: it is present in both mice and humans. GP2- specific antibodies (Anti-GP2-antibodies) developed and provided by MBL has been widely used in research of mucosal immunity. Through elucidation of the antigen-transportation system by the M cells, it is expected that anti- GP2 antibody will promote the development of oral vaccines for infections and cancer, and to contribute to treatment of inflammatory bowel diseases by artificially controlling immunological response of the intestine.

Bad cholesterol and PCSK9

pcsk9LDL cholesterol (LDL-c), referred to as "bad cholesterol," plays an important role in transportation of lipids, which are essential for building up cells in the body, from the liver to peripheral tissues under normal conditions. LDL-c synthesized in the liver is carried by the blood flow and binds to LDL receptors on the surface of cells in peripheral tissues, and the LDL-c/LDL receptor complex is incorporated into cells and used for synthesis of a cell membrane composed of lipids, of fat-soluble vitamins or hormones. However, when blood LDL-c concentration becomes higher than necessary, LDL cholesterolemia, one of the hyperlipidemia, may occur.

In 2003, a mutation in the Proprotein convertase subtilisin/kexin 9 (PCSK9) gene was reported in a family with hereditary LDL cholesterolemia. Functions of PCSK9 have been revealed as follows: (1) it is secreted into blood to bind to LDL receptors on the surface of cells, (2) this binding promotes degradation of LDL receptors and reduces their numbers, and (3) as a result, the uptake of blood LDL-c to cells is reduced with increased LDL-c concentration in blood. The increase of blood PCSK9 itself may cause a risk of occurrence of LDL cholesterolemia.

MBL provides reagents for quantifying PCSK9, as well as kits, related antibodies, and recombinant proteins for research in binding characteristics between PCSK9 and LDLR or in vitro evaluation of inhibitors.

Assay systems for the efficient screening of new drug substance candidates

Based on an assay system that can be used to efficiently explore new drug substance candidates (Drug Screening System), MBL provides activity measuring kits for phosphoenzymes, deacetylases, and phosphatases, which have been developed by CycLex Co, Ltd. Among these enzymes, histone deacetylase (HDAC) has a particularly important role to regulate various genes by changing nucleosome structures. In addition, HDAC is involved in cell cycle progression and differentiation, and it has been reported that the disturbed regulation of these phenomena by HDAC is associated with occurrence of some types of cancer. HDAC inhibitors are expected to serve as anti-cancer drugs, because they could inhibit cell proliferation and induce the final differentiation to reduce an enlargement of tumor in a mouse model. We offer a wide selection of kit including HDACs Deacetylase Fluorometric Assay Kit and SIRT1/Sir2 Deacetylase Fluorometric Assay Kit.
